Abstract | During progression from tumour growth to metastasis, specific integrin signals enable
cancer cells to detach from neighbouring cells, re-orientate their polarity during migration, and
survive and proliferate in foreign microenvironments. There is increasing evidence that certain
integrins associate with receptor tyrosine kinases (RTKs) to activate signalling pathways that are
necessary for tumour invasion and metastasis. The effect of these integrins might be especially
important in cancer cells that have activating mutations, or amplifications, of the genes that
encode these RTKs.

  生物通2006年8月15日报道   来自纪念Sloan-Kettering癌症研究中心的华人科学家郭文军(Wenjun Guo)以及康奈尔大学等研究机构的同事发现β4整合素能够放大ErbB2信号途径,从而促进乳腺肿瘤的发生。研究的结果刊登在8月10日的Cell杂志上。

  ErbB2基因编码一种受体酪氨酸激酶,ErbB2在恶性乳腺肿瘤中通常都发生了扩增,并且与差的预后有关。到目前为止,ErbB2介导的乳腺肿瘤发展机制还没有被完全确定出来。

  为了分析这个信号途径和细胞附着受体(cell-adhesion receptor)整合素β4在ErbB2介导的肿瘤发生过程中的作用,这个研究组将ErbB2诱导的乳腺肿瘤小鼠模型定向删除了β4信号结构域。结果发现,丧失了β4信号途径能抑制乳腺肿瘤的发生和侵入生长。另外的实验证实β4能与ErbB2形成一个复合体并促进转录因子STAT3和c-Jun的活化。STAT3能促进上皮粘附状性和极性的破坏,而c-Jun则是过度增殖所必须的因子。

  最终,β4信号区域的删除提高了ErbB2靶向治疗的效果。这些结果表明β4整合素能够通过放大ErbB2信号途径来促进肿瘤的进级,并且还确定出β4是乳腺癌分子治疗的一个很有潜力的靶标。

是一个人的。这个研究所非常牛的。不过作者的网页上面现在找不到他,估计离开这个实验室了。

Author biographies

Wenjun Guo received a B.Sc. from Nankai University and an M.Sc. from Fudan University in China. He is currently pursuing his Ph.D. under the guidance of Filippo Giancotti in the Sloan-Kettering Division of Weill Graduate School of Medical Sciences of Cornell University, USA. He is studying the role of integrin alpha6beta4 signalling in mammary tumorigenesis by molecular-genetics methods.

Filippo G. Giancotti received his M.D. and Ph.D. degrees from the University of Torino, Italy, and his Board Certification in Hematology and Oncology from the University of Rome, Italy. He received postdoctoral training at the Burnham Institute in La Jolla, California, USA, with Erkki Ruoslahti, and then joined the Department of Pathology of New York University School of Medicine as an assistant professor. Since 1996, he has headed the Laboratory of Cell Adhesion and Signaling at the Sloan-Kettering Institute for Cancer Research of Memorial Sloan-Kettering Cancer Center in New York, USA. Since 2000, he has been a member of the Cell Biology Program of Sloan-Kettering Institute and a professor of Cell Biology and Genetics in the Sloan-Kettering Division of Weill Graduate School of Medical Sciences of Cornell University. The Giancotti laboratory studies the mechanisms by which integrins and other adhesion receptors control cell fate and the contributions of adhesion signalling to development and cancer.

虽然都是周刊,但周刊与周刊差距实在太大了,就上述两个差距也够大的

PNAS, JBC, JACS, NATURE等也是周刊,前面的3本很厚。
